Mission and programs · 2025

Mission

Cancer Prevention Initiative, Inc. (CPI) is a nonprofit corporation whose mission is to accelerate the discovery and development of new medicines that prevent cancer. CPI uses its research, medical and pharmaceutical expertise to invest in first-rate science that will discover new medical methods for preventing cancer. Our strategy is to support research on prevention of inherited cancers as a path towards elimination of all cancers. CPI acts as a facilitator to ensure that good ideas can find sufficient funding to turn them into therapeutic solutions such as drugs or vaccines to prevent cancer. CPI provides financial support, guidance and expertise throughout the research and development process to help researchers use the resources (donations) for the highest possible impact.

Activities or mission (Part I)

Cancer Prevention Initiative, Inc. (CPI) is a nonprofit corporation whose mission is to accelerate the discovery and development of new medicines that prevent cancer. We support research on prevention of inherited cancers as a path toward the elimination of all cancers.

  • In 2025, Cancer Prevention Initiative (CPI) supported five active projects studying therapeutic prevention of inherited cancer. Four of the projects study BRCA1/2 mutated cancers and one project studies Lynch Syndrome. Two of the projects, one at Cleveland Clinic and one at Dana-Farber, are developing immunoprevention strategies to combat BRCA mutant breast cancer. Another immunoprevention project at MD Anderson is developing vaccines to prevent Lynch syndrome-associated colorectal cancer. The project at Harvard/Beth Israel Deaconess Medical center is identifying risk factors in the fallopian tube that gives rise to ovarian cancer in BRCA1 mutation carriers. This work would help develop tests and assays for early detection and risk assessment of ovarian cancer. At Memorial Sloan Kettering Cancer Center, CPI supports a project to prevent Loss of Heterozygosity (LoH) of BRCA genes, a major driver in cancer initiation, and thereby prevent the development of cancer.

    Expenses $731,546 · Grants $725,000

  • Education and engagement programs: As we move closer to our goal of eradicating all cancers through preventative medicines, we believe that raising awareness about inherited cancer syndromes will inspire more individuals to take proactive, life-saving preventive measures. One of the goals is to enhance the scientific reach of the organization and raise awareness on cancer prevention to a broader audience of patients, advocates and donors. Updates on scientific and medical research advances, recent clinical trials, and interviews with experts in cancer prevention are published on web-based platforms.

Compensation is as filed in Part VII: reportable pay from this organization and from related organizations, other compensation, and hours per week. The total, marked calculated, is our sum of the three dollar columns, with a blank column counted as $0; the form itself has no total. On a Form 990-EZ the columns are Part IV's instead, and on a Form 990-PF Part VIII's — compensation, contributions to benefit plans, and expense account — and the total sums those.

Grants on this return

  • University of Texas MD Anderson Cancer Center

    $450,000

The purpose of the Grant is for the Principal Investigator to carry out the research to develop immune interventions that could reduce the risk of Lynch Syndrome (LS)-related cancers. The goals of the project are to develop vaccines and test their potential to prevent colorectal cancer in a preclinical model of LS and to explore the colonic immune microenvironment.

  • This project to identify factors that influence the Loss of Heterozygosity (LOH) was initially funded in 2019. Based on the significant progress made in the project, the project was granted additional funding to continue to phase 2 in 2021 and phase 3 in 2024-2025. During phase 3, they will study allele loss under BRCA wildtype and heterozygous conditions. These studies will provide a comparative mechanistic understanding of LOH at BRCA under normal and carrier conditions. This mechanistic understanding will facilitate development of strategies to prevent LOH and thereby prevent cancer development in high-risk individuals.

  • Scientific and administrative oversight of all Cancer Prevention Initiative programs.
